Description: Activation of tumor necrosis factor receptor-1 (TNFR1; MIM 191190) mediates both proinflammatory and proapoptotic effects. Proapoptotic effects triggered by TNFR1 are dependent on the presence of an 80-amino acid cytoplasmic death domain that recruits other death domain adaptor proteins, such as TRADD (MIM 603500), FADD (MIM 602457), and RIPK1 (MIM 603453), leading to the activation of caspases and the disassembly of cell structures. Proinflammatory and antiapoptotic effects are mediated by the activation of nuclear factor kappa-B (NFKB; see MIM 164011), which leads to the induction of proinflammatory cytokines or to the induction of inhibitors of apoptosis.[supplied by OMIM]
