Exciting and novel preliminary data from our programs demonstrate that (i) reduced expression of heme oxygenase 1 (HO-1) is responsible for loss of interstitial cells of Cajal and delayed gastric emptying in non-obese diabetic (NOD) mice, (ii) upregulation of (HO-1) reverses delayed gastric emptying in this model, perhaps by generating carbon monoxide (CO), which has anti-apoptotic and cytoprotective actions, and may relax smooth muscle, and (iii) hemin upregulates HO-1 in humans. However, hemin is exorbitant and can only be administered intravenously. A large throughput screening assay uncovered that the histamine H2 receptor antagonist nizatidine and the ACE inhibitor lisinopril upregulate HO-1 in Human Embryonic Kidney (HEK) cells. Hence, this double-blind placebo-controlled study will randomly assign 24 healthy subjects to one of 4 arms, and HO-1 protein activity and concentration will be assessed.'}, 'eligibilityModule': {'sex': 'ALL', 'stdAges': ['ADULT', 'OLDER_ADULT'], 'maximumAge': '65 Years', 'minimumAge': '18 Years', 'healthyVolunteers': True, 'eligibilityCriteria': 'Inclusion criteria:\n\n* Healthy subjects without clinical evidence of significant cardiovascular, gastrointestinal, hematological, neurological, psychiatric or other disease that may interfere with the objectives of the study and/or pose safety concerns\n* Normal serum potassium and estimated glomerular filtration rate (eGFR) \\> 60 ml/minute\n* Baseline systolic BP ≥ 110 mmHg\n* No known hypersensitivity to lisinopril or nizatidine\n* Able to provide written informed consent before participating in the study\n* Able to communicate adequately with the investigator and to comply with the requirements for the entire study.\n\nExclusion criteria:\n\n* Pregnant\n* Breast feeding\n* Current smoker\n* Symptoms of functional GI disorder as assessed by a validated questionnaire\n* Previous history of peptic ulcer disease.'}, 'identificationModule': {'nctId': 'NCT02232308', 'briefTitle': 'Modulation of Heme Oxygenase 1 by Nizatidine and Lisinopril in Healthy Subjects', 'organization': {'class': 'OTHER', 'fullName': 'Mayo Clinic'}, 'officialTitle': 'Modulation of Heme Oxygenase 1 by Nizatidine and Lisinopril in Healthy Subjects', 'orgStudyIdInfo': {'id': '14-004137'}, 'secondaryIdInfos': [{'id': 'UL1RR024150', 'link': 'https://reporter.nih.gov/quickSearch/UL1RR024150', 'type': 'NIH'}, {'id': 'P01DK068055', 'link': 'https://reporter.nih.gov/quickSearch/P01DK068055', 'type': 'NIH'}]}, 'armsInterventionsModule': {'armGroups': [{'type': 'EXPERIMENTAL', 'label': 'Nizatidine', 'description': 'Nizatidine (150 mg) will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'interventionNames': ['Drug: Nizatidine']}, {'type': 'EXPERIMENTAL', 'label': 'Lisinopril', 'description': 'Lisinopril (10 mg) will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'interventionNames': ['Drug: Lisinopril']}, {'type': 'EXPERIMENTAL', 'label': 'Nizatidine plus Lisinopril', 'description': 'Nizatidine (150 mg) and Lisinopril (10 mg) will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'interventionNames': ['Drug: Nizatidine', 'Drug: Lisinopril']}, {'type': 'PLACEBO_COMPARATOR', 'label': 'Placebo', 'description': 'Placebo capsules to match active drug will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'interventionNames': ['Drug: Placebo']}], 'interventions': [{'name': 'Nizatidine', 'type': 'DRUG', 'otherNames': ['Axid'], 'description': 'Nizatidine (150 mg) will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'armGroupLabels': ['Nizatidine', 'Nizatidine plus Lisinopril']}, {'name': 'Lisinopril', 'type': 'DRUG', 'otherNames': ['Zestril', 'Prinivil'], 'description': 'Lisinopril (10 mg) will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'armGroupLabels': ['Lisinopril', 'Nizatidine plus Lisinopril']}, {'name': 'Placebo', 'type': 'DRUG', 'description': 'Placebo capsules to match active drug will be administered once daily for the first 3 days, then twice daily for days 4-9, and once on day 10.', 'armGroupLabels': ['Placebo']}]}, 'contactsLocationsModule': {'locations': [{'zip': '55905', 'city': 'Rochester', 'state': 'Minnesota', 'country': 'United States', 'facility': 'Mayo Clinic in Rochester', 'geoPoint': {'lat': 44.02163, 'lon': -92.4699}}, {'zip': '55905', 'city': 'Rochester', 'state': 'Minnesota', 'country': 'United States', 'facility': 'Mayo Clinic', 'geoPoint': {'lat': 44.02163, 'lon': -92.4699}}], 'overallOfficials': [{'name': 'Adil Bharucha, MBBS, MD', 'role': 'PRINCIPAL_INVESTIGATOR', 'affiliation': 'Mayo Clinic'}]}, 'sponsorCollaboratorsModule': {'leadSponsor': {'name': 'Adil Bharucha', 'class': 'OTHER'}, 'collaborators': [{'name': 'National Center for Research Resources (NCRR)', 'class': 'NIH'}, {'name': 'National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK)', 'class': 'NIH'}], 'responsibleParty': {'type': 'SPONSOR_INVESTIGATOR', 'investigatorTitle': 'Professor of Medicine', 'investigatorFullName': 'Adil Bharucha', 'investigatorAffiliation': 'Mayo Clinic'}}}}
