ADT is an important part of treatment, because it improves survival for men with metastatic or high-risk localized disease, and reduces rates of biochemical progression for men with intermediate-risk localized disease who receive radiation. The most common ADT agents modulate gonadotropin-releasing hormone to suppress the downstream testosterone production, resulting in testosterone levels similar to those observed following surgical castration (\\<20 ng/dL). Since male sexual interest is highly correlated with serum testosterone levels, loss of sexual interest is nearly universal among men who receive ADT.Sexual dysfunction is the most common complaint among men with prostate cancer and contributes to lower overall quality of life (QoL) experienced by men receiving ADT. Furthermore, the loss of sexual interest experienced during ADT is highly distressing for men with prostate cancer and their partners, which contributes additional psychological morbidity in these patients.\n\nFlibanserin is approved for treatment of female hypoactive sexual desire disorder, and the safety profile of 100mg daily flibanserin is well described in premenopausal women.The safety profile of flibanserin in healthy men has been assessed in multiple phase I clinical trials, but has not been evaluated among men receiving ADT for prostate cancer.\n\nThis is a phase II randomized, double-blinded, placebo-controlled clinical trial designed to provide an initial estimate of the efficacy of flibanserin to promote sexual interest in men with prostate cancer receiving androgen suppression therapy and to confirm the safety profile. This study will take place at a single academic comprehensive cancer center.\n\nFollowing confirmation of eligibility, participants who are enrolled in this study are randomized to receive daily flibanserin 100mg or placebo for a 12-week period.'}, 'eligibilityModule': {'sex': 'MALE', 'stdAges': ['ADULT', 'OLDER_ADULT'], 'minimumAge': '18 Years', 'healthyVolunteers': False, 'eligibilityCriteria': 'Inclusion Criteria:\n\n* Provision of signed and dated informed consent form.\n* Stated willingness to comply with all study procedures and availability for the duration of the study.\n* Ability to take oral medication and be willing to adhere to the study regimen.\n* Male age \\>18 years.\n* Histologically confirmed prostate cancer.\n* Currently receiving gonadotropin releasing hormone agonist/antagonist monotherapy.\n* Serum testosterone \\<50 ng/dL.\n* Serum AST and ALT less than 2 times upper limit of normal.\n* Endorsed reduced sexual interest.\n* Attempted intercourse.\n* Current sexual partner.\n* Was sexually active with partner within 6 months prior to ADT.\n* No other antineoplastic therapy planned during study period.\n* No active symptoms attributable to systemic prostate cancer.\n\nExclusion Criteria:\n\n* Current systemic prostate cancer treatment besides GnRH agonist/antagonist, anti-androgens, or abiraterone.\n* Prior to ADT had erections not firm enough for intercourse despite use of pharmacologic agents such as phosphodiesterase-5 inhibitors.\n* Current symptoms attributable to active prostate cancer\n* Moderate or heavy alcohol use (\\>2 drinks/day)\n* Concurrent moderate or strong CYP3A4 inhibitors\n* Concurrently taking medication classified as a monoamine oxidase inhibitor.'}, 'identificationModule': {'nctId': 'NCT04743934', 'acronym': 'RAD 2003', 'briefTitle': 'Flibanserin in Men Receiving Androgen Suppression for Prostate Cancer', 'organization': {'class': 'OTHER', 'fullName': 'University of Alabama at Birmingham'}, 'officialTitle': 'RAD 2003/XUAB2104: Randomized Phase II Trial of Flibanserin in Men Receiving Androgen Suppression for Prostate Cancer', 'orgStudyIdInfo': {'id': 'IRB-300006880'}, 'secondaryIdInfos': [{'id': '5R21CA259808-02', 'link': 'https://reporter.nih.gov/quickSearch/5R21CA259808-02', 'type': 'NIH'}]}, 'armsInterventionsModule': {'armGroups': [{'type': 'EXPERIMENTAL', 'label': 'Flibanserin + ADT', 'description': 'Flibanserin at 100mg by mouth once daily at bedtime while receiving androgen deprivation therapy (ADT).', 'interventionNames': ['Drug: Flibanserin 100 MG', 'Drug: Androgen deprivation therapy']}, {'type': 'PLACEBO_COMPARATOR', 'label': 'Placebo + ADT', 'description': 'Placebo at 100mg by mouth once daily at bedtime while receiving androgen deprivation therapy (ADT).', 'interventionNames': ['Drug: Placebo', 'Drug: Androgen deprivation therapy']}], 'interventions': [{'name': 'Flibanserin 100 MG', 'type': 'DRUG', 'description': 'Flibanserin 100mg tablets taken by mouth daily at bedtime', 'armGroupLabels': ['Flibanserin + ADT']}, {'name': 'Placebo', 'type': 'DRUG', 'description': 'Visually identical placebo tablets taken by mouth daily at bedine', 'armGroupLabels': ['Placebo + ADT']}, {'name': 'Androgen deprivation therapy', 'type': 'DRUG', 'description': 'Androgen deprivation therapy consisting of a GnRH agonist or antagonist, choice of agent at discretion of treating physician.', 'armGroupLabels': ['Flibanserin + ADT', 'Placebo + ADT']}]}, 'contactsLocationsModule': {'locations': [{'zip': '35233', 'city': 'Birmingham', 'state': 'Alabama', 'country': 'United States', 'facility': 'University of Alabama at Birmingham', 'geoPoint': {'lat': 33.52066, 'lon': -86.80249}}], 'overallOfficials': [{'name': 'Andrew McDonald, MD', 'role': 'PRINCIPAL_INVESTIGATOR', 'affiliation': 'University of Alabama at Birmingham (UAB)'}]}, 'ipdSharingStatementModule': {'ipdSharing': 'UNDECIDED'}, 'sponsorCollaboratorsModule': {'leadSponsor': {'name': 'Andrew McDonald', 'class': 'OTHER'}, 'collaborators': [{'name': 'National Cancer Institute (NCI)', 'class': 'NIH'}], 'responsibleParty': {'type': 'SPONSOR_INVESTIGATOR', 'investigatorTitle': 'Principal Investigator', 'investigatorFullName': 'Andrew McDonald', 'investigatorAffiliation': 'University of Alabama at Birmingham'}}}}
